Caspase-3 is an effector caspase. The assay is based on the specific peptide substrate DEVD-pNA (Asp-Glu-Val-Asp-p-nitroanilide) hydrolysis by caspase-3.
Caspase-3 is the primary terminal protease in apoptosis and the most widely studied caspase. It activates pro-caspase-2, 6, 7, and 9, specifically hydrolyzing several key apoptotic proteins, such as PARP, mediating chromatin condensation, apoptotic body formation, and nuclear DNA fragmentation. The assay is based on the specific hydrolysis of the peptide substrate DEVD-pNA (Asp-Glu-Val-Asp-p-nitroanilide) by caspase-3. The released free nitroanilide pNA has a maximum absorbance at 405 nm.
Caspase-3 Colorimetric Assay Kit provides a convenient and simple way for detecting the DEVD-dependent caspase activity. Since DEVD-pNA is also a substrate for caspase-7, this product can be used to detect caspase 3/7 activity.
