Caspase-6, also known as Mch-2, can induce apoptosis. Caspase-6 specifically hydrolyzes the peptide substrate VEID-pNA (Val-Glu-Ile-Asp-p-nitroanilide).
Caspases are a family of proteases involved in apoptosis, encompassing over ten members. Caspase-6, also known as Mch-2, forms an activated caspase-6 dimer after its precursor protein is cleaved by granzyme B, which can induce apoptosis. Caspase-6 cleaves PARP, a key regulator of apoptosis, and Lamin A, a key protein on the nuclear membrane. Its recognition site for Lamin A is VEID. Assay principle: Caspase-6 specifically hydrolyzes the peptide substrate VEID-pNA (Val-Glu-Ile-Asp-p-nitroanilide), releasing free nitroanilide pNA with a maximum absorbance at 405 nm.
