BrdU is a synthetic thymidine analog, non-endogenously expressed in tissue cells and relatively widely used for the detection of cellular DNA synthesis.
Currently, four cell proliferation markers are commonly used: DNA polymerase α (DNA polymerase α), proliferating cell nuclear antigen (PCNA), Ki-67, and 5-bromodeoxyuridine (Brdu). BrdU, also known as bromide-deoxyuridine, is a synthetic thymidine analog that selectively incorporates into cellular DNA during S phase, replacing thymidine. This allows for the detection of cellular DNA synthesis and the marking of cell division, apoptosis, and other behaviors. In genetic studies, BrdU can be injected into cells in vivo or loaded into cell culture. Anti-BrdU monoclonal antibodies can then be used for specific labeling, and proliferating cells can be visualized using ICC or IHC staining. Furthermore, dual staining can be combined with other cell markers to identify the type and rate of proliferating cells, making it crucial for studying cell dynamics.
